Cinnamomum zeylanicum Blume, known for its medicinal and culinary value, was analyzed for comparative phytochemical profiling and antioxidant potential between Indian (Shillong, Kolkata) and African (Tanzania) stem bark varieties. Using HPTLC and GC-MS, the essential oils revealed key variations in chemical constituents, notably the exclusive presence of eugenol in the Tanzanian variety and higher cinnamaldehyde dimethyl acetal content. Physicochemical and organoleptic differences reflected geographic influence. Antioxidant studies using DPPH and FRAP assays confirmed superior activity in the Tanzanian sample, with the lowest IC50 (22.05 µg/ml) and highest FRAP value (579 µM). Phytochemical screening confirmed the presence of multiple bioactive compounds in the samples. These results underscore the significance of geographical origin in the quality and efficacy of medicinal plants, supporting the need for standardization protocols. This study provides a robust framework for evaluating regional variations in C. zeylanicum, enhancing its pharmacological validation and ensuring authenticity in herbal formulations. Molecular docking study with eugenol revealed strong binding affinity of eugenol with protein targets PTP1B, PPARγ, PPARδ, and PPARα in diabetes, and with BACE1 in Alzheimer's disease. Show less

Liver steatosis, fibroinflammation, and iron overload, are growing global health concerns, yet the genetic architecture and causal pathways linking liver pathology to systemic disease remain incompletely understood. We analysed MRI-derived liver traits—corrected T1 (cT1), proton density fat fraction (PDFF), and liver iron—in 37,626 UK Biobank participants. Genome-wide (GWAS), transcriptome-wide (TWAS), and GWAS identified 18 loci for cT1, 15 for PDFF, and 5 for liver iron, including six not previously reported. TWAS, This integrative imaging-genetics study reveals 13 potentially novel genes and several protein candidates implicated in hepatic steatosis, inflammation, and iron homeostasis. These findings enhance understanding of liver disease biology and may help identify new targets for early detection or treatment. This large imaging-genetics study in over 37,000 people identifies genetic and protein factors linked to liver fat, fibroinflammation, and iron levels. It shows that higher liver fat and inflammation are associated with increased cardiometabolic risk, while higher liver iron appears inversely linked to risk of heart disease. Community structures are common features of many real-world networks, and community detection is necessary to understand how these networks are organized. Various approaches have been devised for community detection, with each providing varying degrees of both accuracy and structural understanding. One of them, the Label Propagation Algorithm, is so common because it is simple and computationally cheap. Nevertheless, it does not usually reach great modularity and yields inaccurate community counts and structures in real-world networks. This is mostly due to its naive criteria of selecting the neighbor nodes when it comes to label propagation. To tackle the issue, we developed an adjusted algorithm, which we call Embedding-based Label Propagation (ELP), a hybrid between LPA and node embedding that allows us to combine both local connectivity and global structural data. ELP update step takes into consideration not only the local neighborhood, as in conventional LPA, but also embedding-based similarities to inform more productive neighbor selection. We tested ELP on popular benchmark datasets such as Karate Club, Dolphins, Football, Polbooks, and LFR synthetic networks and compared its results with LPA and other well-established algorithms. The empirical findings show that ELP can always perform better in modularity, NMI and NF1 scores, but it is also scalable to large and complex networks. These results can be used to identify ELP as an effective and powerful method of community-finding in real and artificial-world scenarios. Show less

Atherosclerosis is a chronic condition characterized by impaired lipid homeostasis and chronic inflammatory pathology in large and mid-sized arteries. Myocardial infarction is caused by coronary artery thrombosis in a ruptured or unstable atherosclerotic plaque. Despite the emphasis on known triggering factors, such as hypertension and dyslipidemia, adverse events following MI, such as recurrence and mortality, are still high. Therefore, it is imperative to assess potential determinants of plaque instability. We evaluated markers of inflammation, extracellular matrix (ECM) remodeling, thrombosis, and lipids in first-time and recurrent MI (RMI). Two hundred patients diagnosed with MI within the first 24 h of the event were included in the study and categorized as first-time or recurrent MI. Serum levels of NF-κB, hs-CRP, TNF-α, IFN γ, IL-6, VCAM-1,MMP-9, stromelysin, TIMP-1, MCP-1, PAPP-A, vWF, D-dimer, PLA2, PON-1, Apo-B, Apo-A1, ox-LDL, and anti-oxidized LDL antibodies were analyzed by ELISA. We performed a multivariate logistic regression analysis for risk stratification. The mean age of first-time MI patients was 52.4 ± 25 years and that of recurrent MI patients was 55.9 ± 24.6 years. Obesity plays a pivotal role in the development of metabolic syndrome-excessive body fat, spikes in blood glucose levels and hypertension-and ultimately leads to cardiovascular diseases and type 2 diabetes (T2D), if left unattended. The present study aimed to investigate the associated risk of T2D with obesity risk alleles of fat mass and obesity-associated (FTO) and melanocortin 4 receptor (MC4R) genes. The study includes 400 subjects (300 T2D diabetic cases and 100 healthy controls). Genetic analysis was done by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods. The findings of the study show no significant increase in odds of diabetes associated with the prevalence of FTO and MC4R minor alleles. Rare allele frequencies for "A" of FTO rs9939609 were 0.34 and 0.30 in cases and controls, respectively. Rare allele frequencies for A of MC4R rs12970134 were found to be more common in controls (0.45) than cases (0.41), but the difference was insignificant (p 0.246); however, an increase in body weight with the presence of allele "A" of the FTO gene (p value < 0.001) was found, indicating indirect involvement in the development of T2D. In addition, these were also correlated with the demographic/lifestyle and clinico-pathological parameters between T2D cases and controls. We found that T2D patients with a history of smoking and high consumption of alcohol, fast foods and sweetened beverages are at high risk of T2D compared to healthy controls (p  < 0.01*). The present study concludes that there is no direct association of rs9939609 of the FTO gene with the occurrence of diabetes in the Indian population, but its role in T2D development cannot be overlooked altogether. Furthermore, we conclude that the rs9939609 of FTO carries a potential risk of obesity and because of this FTO rs9939609 T > A is widely considered an obesity-associated allele/single-nucleotide polymorphism (SNP).
